Curative effect mechanism of Xianlian Jiedu decoction in treatment of patients with stage Ⅲ colorectal cancer based on transcriptomics

Abstract:

Objective

To explore the potential mechanism of Xianlian Jiedu decoction (XLJDD) in the treatment of stage Ⅲ colorectal cancer patients via transcriptome sequencing technology.

Methods

This study focused on patients with stage Ⅲ colorectal cancer. Four tumor tissue samples were randomly selected from the XLJDD group and four from the control group who did not receive XLJDD. Full transcriptome sequencing was performed to identify differentially expressed genes, enrich functional pathway, and construct a miRNA-mRNA regulatory network.

Results

By comparing samples of the two groups, it was found that there were multiple differentially expressed miRNA and mRNA in the samples of colorectal cancer patients taking XLJDD, including hsa-miR-105-5p, hsa-miR-129-1-3p and hsa-miR-135a-5p, as well as ANKRD31, ESPNL and DACT2. Functional enrichment analysis showed that membrane transport function, steroid hormone response, IL-4, IL-13 and other tumor-related pathways were significantly enriched after XLJDD treatment.

Conclusion

XLJDD may interfere with the tumor progression of stage Ⅲ colorectal cancer patients by regulating the expression levels of miRNA and mRNA and affecting related pathways.
