Preliminary experimental study on the expression of α1 adrenergic receptor subtypes in human liver sinusoidal endothelial cells

Abstract:

Objective

To investigate the expressions of α1 adrenergic receptor (α1AR) subtypes in liver sinusoidal endothelial cells (LSEC).

Methods

Human primary LSEC were cultured in vitro. The effects of α1AR agonists phenylephrine and α1AR blocker doxazosin on the expressions of angiogenesis-related markers CD31 and vWF in LSEC and the expressions of various α1AR subtypes in LSEC were verified by Western blotting analysis. Immunohistochemistry was used to verify the expressions of α1AR subtypes in mouse liver.

Results

α1AR agonist phenylephrine could increase the expressions of CD31 and vWF in LSEC, and α1AR blocker doxazosin antagonized this effect. The expressions of α1aAR and α1bAR were detected in LSEC in vivo and in vitro, but the expression of α1dAR was not detected.

Conclusions

α1AR is involved in the regulation of LSEC phenotype, and this function is achieved through two subtypes of α1aAR and α1bAR. Therefore, α1AR may become a new regulatory target of liver fibrosis.
